If still no go, try running the defragmenter in Safe Mode.
YOU RULE!!!!! I don't intend on using the the window's defrag but I'm concerned as to why it isn't there. I then ran the seearch for Disk Defragmenter and nothing showed up. Reinstall Disk Defragmenter Windows 10 YOU RULE!!!!!
After allowing/encouraging this to happen on start-up, C reappeared in the Disk Defrag selection menu. I copied the one from the ServicePackFiles\i386 folder over, and restarted the Defragmenter, and it worked!

Shane. You are the greatest! Anyone getting the "Disk Defragmenter Could not start" message and on Windows XP Home This is what worked for me. Windows Unknown Major Release Service Pack 0 Professional X86 64-bit Note: Some versions of Win 7 disable the ability to run the built in defragger in safe mode.
